Agenerase
What is it?
A retroviral, protease (enzyme) inhibitor available as capsules in two strengths, containing 50mg or 150mg amprenavir. Also, AGENERASE ORAL SOLUTION containing 15mg amprenavir per ml.
Medical Uses
Combined with other retroviral drugs to slow progression of disease in H1V infected patients.
Dosage
Adults, tablets; people weighing less than 50kg: 20mg per kg body weight, twice each day. Maximum dose, 2400mg. People weighing more than 50kg: 1200mg twice each day. If drug is being combined with ritonavir, take 600mg twice each day. Children aged 4 and over: 20mg per kg of body weight twice each day; maximum daily dose, 2400mg. Agenerase oral solution; Adults: 1.1mg per kg of body weight three times each day; maximum daily dose, 2800mg. Children aged 4 and over: 1.1mg per kg of body weight three times each day; maximum daily dose, 2800mg.
Special Care
Pregnancy, patients with liver disease.
Avoid Use
Children aged less than 4 years, breastfeeding, liver or kidney impairment (if using oral solution).
Possible Drug and Food Interactions
Oral contraceptives, St John's Wort, antacids, cisapride, triazolam, flurazepam, terfenadine, diazepam, pimozide, astemizole, midazolam, CYP3A4 substrates, ergot derivatives, phenytoin, rifampicin, amiodarone, lignocaine, phenobarbitone, warfarin, methadone, TCADs, quinidine.
Known Side Effects
Gastrointestinal upset, rash or allergic symptoms (discontinue). Any adverse side effects should be reported to the CSM.
