What is it?

A dopamine agonist, pergolide mesylate, produced as tablets: ivory, marked 4131 contain 0.05mg, green marked 4133 (0.25mg), and pink tablets, marked 4135 (1mg). All are rectangular, scored and marked LILLY.

Medical Uses

Sole therapy or additional therapy (to levodopa) in treatment of Parkinson's disease. Dosage : additional therapy, 0.05mg each day at first in 3 divided doses increasing every third day by 0.1-0.15mg for a period of 12 days. Then the dose is increased by 0.25mg every third day until the best response is achieved. Maximum daily dose is 5mg. Levodopa dose should be carefully reduced. Sole therapy, at first, 0.05mg taken as evening dose on day one then 0.05mg taken at noon and evening on 2nd to 4th days. Afterwards, dose is gradually increased over 25 day period to 1.5mg each day in 3 divided doses, morning, noon and evening. Dose may then be further increased, if needed, by 0.25mg twice each week until optimal response is achieved.

Special Care

Pregnancy, breast-feeding, heart disease, heart arrhythmias, serosal inflammatory disorders associated with ergot derivatives, history of hallucinations. Patient should be monitored for signs of fibrosis. Drug should be gradually withdrawn.

Avoid Use

Children.

Possible Drug and Food Interactions

Anticoagulants, antihypertensives, other dopamine antagonists, drugs that affect protein binding.

Known Side Effects

Disturbances of heartbeat, movement disorder (dyskinesia), drowsiness, hypotension, inflammation of the nose, dyspepsia, nausea, dyspnoea (laboured breathing), diplopia, gastrointestinal upset, abdominal pain, abnormal liver tests and heart beat rate, rash, fever, derosal inflammatory disorders.
