What is it?

A vitamin K derivative available in 4 different forms: sugarcoated, white tablets contain 10mg of phytomenadione; KONAKION MM, available as a solution in ampoules for injection containing 10mg of phytomenadione per ml in mixed micelles; KONAKION NEONATAL, available in ampoules for injection containing 1mg of phtomenadione per 0.5ml; KONLM1ON MM PAEDIATRIC, available as a solution containing 2mg of phytomenadione per 0.2ml in mixed micelles.

Medical Uses

As an antidote to hypoprothrombinaemia (an abnormally low level of prothrombin clotting factor II in the blood, characterized by bleeding and loss of blood clotting ability). Konakion Neonatal, is Also, used for prevention of haemorrhagic disease of the newborn (a bleeding disorder of newborn infants caused by vitamin K deficiency), in those born at or after 36 weeks gestation. KoniakionMM Paediatric, prevention and treatment of haemorrhagic disease in newborn babies.

Dosage

Adults, tablets, 5 to 20mg either allowed to dissolve in mouth or chewed. KoniaionMM, 10 to 20mg given over 3 to 5 minutes by slow intravenous injection or by infusion intravenously, over 20 minutes to 1/z an hour. This is given after the anticoagulant has been stopped. After 3hours, prothrombin levels are measured and the dose may then be repeated, to a maximum of 40mg in 24 hours, if the response has not been satisfactory. Children, aged over 3 months, tablets, 5 to 10mg dissolved in mouth or chewed. Konakion Neonatal, as an antidote, as directed by specialist haematologist; prevention of haemorrhagic disease, 1mg given by intramuscular injection at birth or as soon as possible; neonatal preparation should be used in all infants aged under 12 months. KonakionMM, limited information is available and should only be used in children under expert supervision; the following dosages do not apply to children who cannot have warfarin withdrawn. Children aged 12 months to 18 years, for haemorrhage in hypoprothrombinaemia, 2 to 5mg, either by intravenous infusion or slow intravenous injection. To lessen risk of bleeding, 1 to 5mg by infusion or slow intravenous injection. Prothrombin levels should be assessed after 2 to 6 hours and the dose may then be repeated if response is not satisfactory. KonakionMM Paediatric, prevention of haemorrhagic disease, 2mg given by mouth at birth followed by further 2mg between 4 and 7 days old. Breast- fed babies should receive further 2mg at 1 month old and then every month, as advised, unless formula milk is introduced. Treatment of haemorrhagic disease, 1mg given intravenously at first and further doses under specialist supervision, consulting manufacturer's instructions. For premature babies and those at special risk, manufacturer's instructions should be consulted. Special cafe: elderly patients.

Known Side Effects

Sweating, flushing, lack of oxygen in blood (cyanosis) causing bluish tinge to skin and mucous membranes. Symptoms of analphylaxis may occur with injection.
