Prograf
What is it?
An immunosupressant and macrolide lactone available as capsules in 3 strengths all containing tacrslimus. Yellow capsules, marked with strength and [f]607 contain 0.5mg; white capsules marked with strength and [f]617 contain 1mg; red- grey capsules marked with [strength and [f]657 contain 5mg. Also, PROGRAF CONCENTRATE FOR INFUSION, available as a concentrated solution in ampoules containing 5mg of tacrolimus per ml.
Medical Uses
Immunosuppression in patients receiving liver or kidney transplants, rejection of transplanted liver or kidney in patients in whom other immunosuppressant regimes have failed.
Dosage
Adults and children, manufacturer's literature must be consulted.
Special Care
Liver disorders. Blood glucose, electrolytes, coagulation and haematology require monitoring - Also, blood pressure and vision. Anti- lymphocyte treatment should not be given at the same time.
Avoid Use
Pregnancy, breastfeeding.
Possible Drug and Food Interactions
Potassium-sparing diuretics, potassium supplements, drugs bound to plasma proteins, cyclosporin, live vaccines, drugs affecting P450, oral contraceptives.
Known Side Effects
Sensory and central nervous system disorders, high blood pressure, headaches, lymphoproliferatiwe, gastrointestinal, haematological and heart and circulatory disorders, infection, high blood glucose levels, enlarged, damaged heart, skin and respiratory disorders, tremor, sensation of numbness/'pins and needles', liver and kidney disorders. Possible anaphlactic reactions to infusion.
